A groundbreaking study recently released in Nature by researchers at the University of Toronto has unveiled the presence of ancient DNA within human genomes that harbors remarkable potential. These genetic elements, which have persisted throughout human evolution, can activate robust immune responses capable of eradicating cancer cells.
Daniel De Carvalho, one of the lead authors of the research, stated: “Throughout millions of years of evolution, humans have accumulated a variety of ‘silent’ repetitive elements in our DNA, but their purpose has remained largely a mystery.”
As "genome archaeologists," the team aimed to uncover the role of these genetic artifacts and discovered that, under specific conditions, they can be reawakened to stimulate our immune defenses.

The DNA remnants referred to in the study are known as SINEs, or Short Interspersed Nuclear Elements. These repetitive sequences typically lie dormant, but they can be activated with the aid of epigenetic drugs.
Pharmaceutical agents that target these elements trigger the production of double-stranded RNA. The body misinterprets this surge of double-stranded RNA as an infection and mobilizes its innate immune defenses, which can include pathways capable of targeting cancer.
Subsection 1.1.1: Challenges in Immune Response
Despite the immune system’s potential to eliminate cancer cells, tumors have developed various tactics to evade detection. One such strategy involves the production of ADAR1, an enzyme that inhibits the activation of SINE-derived double-stranded RNA.
“Given that ADAR1 operates enzymatically, our findings present an exciting new target for developing a novel class of drugs that could harness these ancient defenses within our genome,” remarked De Carvalho. The study revealed that inhibiting the tumor's capacity to produce ADAR1 significantly increased its vulnerability to immune system attacks.
